Our oligos are synthesized on an ABI 3900 or a BioAutomation MerMade-6 synthesizer using beta-cyanoethyl chemistry. Synthesis begins with the 3' terminal nucleoside that is attached to a solid support, and the remaining nucleotide bases are added one at a time until the synthesis is complete.

Chemical reactions in the synthesis cycle are not flawless, rather 98-99% efficient. With each base addition, small amounts of sequences fail to undergo a base addition or coupling. Most of these failed sequences are chemically capped during the synthesis cycle preventing them from participating in further base additions. However, because efficiency is not 100% some sequences will remain uncapped and are available for further base additions, resulting in n-1-mer, n-2-mer, etc. failure sequences. Reverse phase cartridge purification allows for the separation of the full-length product from these failure sequences.

Purification Considerations:

Yield is length-dependent for RPC purification.

Maximum allowable length for RPC purification is 25 bases for the 40 nmol scale and 50 bases for the 200 nmol and 1 μmol scales.

Synthesis of specialty oligos with minor bases or labels is available through the purchase of the specialty reagent. Once diluted, the shelf life of most of these reagents is 1-2 days making it necessary to charge for the entire bottle. One bottle of reagent is usually sufficient for the synthesis of multiple labeled oligos depending on the synthesis scale (detailed information below), so plan ahead for cost-effectiveness.

Standard Ordering

For your convenience and the fastest turnaround, we offer several options of ordering directly from our lab. No PeopleSoft requisition is required when using any standard ordering method.


By Synthesis Request Form

Typed information is preferred, but forms may also be printed and completed by hand. Completed forms may be hand-carried, e-mailed*, or faxed to the Core Facility. After normal operating hours, forms may be left in the Oligo Drop Box located outside the lab.

To type a form, use the Online Oligo Request Form and either print it or email it as an attachment; to complete a form by hand, use the Oligo Request Form.

By Email Without a Form (1-9 oligos)

Authorized users on an account may email oligo requests to oligos@uthscsa.edu. Please use the following guidelines:

  1. Include all of the following information:
    • USER NAME (first and last name)
    • PI NAME
    • CONTACT PHONE NUMBER
    • ACCOUNT NUMBER
    • DEPARTMENT
  2. Use all CAPS for oligo sequences.
  3. Do not use spaces in oligo sequences.
  4. Indicate scale and level of purification for each oligo.

General Stores Ordering

If standard ordering was used, you will be notified by the NACF staff by phone or email as soon as your order is ready for pick-up. Off-campus users must include shipping information if orders are to be sent directly to off-campus locations. When an order is placed through General Stores, General Stores will contact you by phone when the order is ready. Off-campus orders made through General Stores will be delivered following Stores' policies.

*Important Note: Synthesis services are offered on a first come, first serve basis. This means that when our daily maximum run capacity is reached, oligos will be in queue to be processed. Our maximum run capacity limits us to 45 same day primers, which must be received by the NACF by 9:30am. Every effort is made to maintain quick turnaround times, but unfortunately we cannot guarantee them. Users will be notified at the time of submission or promptly afterward if an expected turnaround time is not going to be met. Off-campus users using campus mail may expect a delayed turnaround time.
